Milestone-based Cryo-EM Services

Milestone 1

Grid Screening

Vitrify sample, capture small number of images representing thickness range, evaluate by eye for optimal particle mono-dispersity, density and morphology, and deliver report on recommended next steps.

Milestone 2

Pilot dataset and processing

Collect ~500 micrograph movies, using the same conditions/setup as a full dataset; process data; and, if 2D processing goes well, proceed to 3D for deeper insight.

Milestone 3

Full dataset collection and processing

To "Collect 5,000 to 15,000 micrograph movies over 24 hours (depending on grid type and collection parameters), and report on findings and expert analysis (final particle count, resolution, FSC curves, particle angular distribution, sharpening and local resolution). Also provide half maps, full map, sharpened map and sharpened map filters to local resolution.

Milestone 4

Atomic model-building

Using either an initial model or directly interpreting the sequence from a high-resolution map, Helix experts deliver a comprehensive Table 1 summary detailing model geometry statistics and map-to-model fit. The final atomic model is provided in both .pdb and .cif formats, ensuring that your data is ready for publication or further analysis.
