Helix's versatile recombinant protein production capabilities include gene construct design, protein expression, and protein purification. Proteins can be produced for any downstream analytical technique, from assays to biophysical and structural characterization, across both biopharmaceutical and agricultural applications. Helix's expertise includes crystallization-grade protein production with purity of 95% or higher.

Construct Design/ Synthesis

Protein design matters. We work with you to ensure each construct sequence is biologically relevant, has appropriate purification tags, and meets requirements for its intended assays or biophysical/structural characterization.

Pilot Expression Testing

Pilot testing proactively fine tunes expression and purification parameters before scale up, saving time and cost. We use small-scale cell culture to test the construct we've designed, looking not only at whether protein expression occurs, but also how much expression will ensure success during scale-up expression and purification. (Optional for clients having well-characterized expression and purification data)

Expression Optimization

When protein expression isn’t optimal, Helix works closely with clients to adjust parameters and increase yield. Techniques include testing under different expression conditions, changing the cell line, optimizing the construct, and/or using an optimal buffer condition that will maintain protein quality.

Scale-up Expression & Purification

Using what is identified during pilot expression testing (or using the client’s data), Helix will produce the amount of protein required for the experiment, from milligrams to grams.

Quantitative Analysis/Quality Control

Each recombinant protein produced includes rigorous quantitative analysis (QA) and quality control (QC). For QA/QC, Helix uses SDS-PAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis), chromatograms, and mass spectrometry to gauge the purity of the protein. Additional analyses of final product concentration include spectrophotometry (UV-Vis), densitometry, and, if required, biophysical characterization to further qualify the protein (i.e. thermal shift assay, DLS, mass photometry, peptide mass fingerprinting).

Throughout the protein production process, project status is communicated weekly. The Helix protein production team provides detailed technical project reports at the end of each stage (time per stage varies depending on which expression system is used).
